Melanotan-2 is a synthetic cyclic heptapeptide designed as an analogue of alpha-melanocyte-stimulating hormone. Its sequence incorporates a lactam bridge that constrains the peptide into a ring, which increases resistance to enzymatic breakdown relative to the natural hormone. Researchers at the University of Arizona synthesised the compound in the late 1980s and early 1990s while studying pigmentation pathways. It has never received marketing approval from any national medicines regulator. In the scientific literature it is usually described as a laboratory research reagent rather than a therapeutic product.
The peptide acts as a non-selective agonist at melanocortin receptors, showing affinity for MC1R, MC3R, MC4R and MC5R. Activation of MC1R on melanocytes drives the conversion of tyrosine into melanin and shifts production toward the darker eumelanin form. MC4R signalling in the central nervous system is linked to appetite and energy balance, which helps explain why reduced food intake appeared in early human studies. Effects on MC4R and on vascular tone also account for the erectile responses recorded as unexpected findings in those same trials.
Melanotan-2 is frequently confused with afamelanotide, a linear analogue authorised in the European Union for erythropoietic protoporphyria. The two compounds differ in chain length, ring structure and receptor selectivity, so findings for one cannot be transferred directly to the other. Published controlled human data on melanotan-2 remain sparse, and much of what circulates online derives from small studies or unpublished reports. Questions about effect size, dose-response behaviour and long-term safety therefore remain unresolved.
The compound was developed in the late 1980s and 1990s by academic researchers investigating photoprotection. The rationale held that stimulating melanin production might reduce ultraviolet damage to skin and lower skin cancer risk. Early work examined receptor binding, pigment response, and short-term tolerability in small studies. That program did not produce an approved drug, and formal development stalled after early-phase trials. Whether induced pigmentation confers meaningful photoprotection remains an open question.
Outside regulated medicine, melanotan II circulates through online vendors as a research chemical, often marketed for tanning. Products sold this way vary widely in purity, concentration, and labeling accuracy, and independent testing has documented discrepancies. Published reports describe both pigment effects and adverse reactions, including nausea, flushing, and darkening of existing moles. Long-term safety data are sparse, and no large controlled trial has established a risk profile. Questions about cumulative effects on melanocytes remain unresolved in the literature.

Structurally, Melanotan-2 retains the core recognition motif of alpha-melanocyte-stimulating hormone while adding a lactam bridge that links two side chains and constrains the molecule into a ring. This modification lowers susceptibility to enzymatic degradation. The compound acts as an agonist at melanocortin receptors, particularly subtypes associated with melanin production. Because the same receptor family influences several physiological processes, researchers note that its activity is not confined to pigmentation alone. Receptor selectivity continues to be examined in published studies.
Melanotan-2 is a synthetic cyclic heptapeptide designed as an analogue of alpha-melanocyte-stimulating hormone, a naturally occurring peptide involved in pigmentation signalling. Its sequence incorporates modified residues that increase potency and extend biological activity relative to the native hormone. The compound binds receptors of the melanocortin family and is examined mainly in laboratory research. It does not occur naturally and exists only as a manufactured chemical entity produced by solid-phase synthesis.
The peptide was developed during the 1980s by researchers investigating melanocortin signalling and skin pigmentation pathways. Early work focused on analogues of alpha-melanocyte-stimulating hormone that would resist enzymatic breakdown more effectively than the parent molecule. Melanotan-2 emerged from that programme as a shortened, cyclised variant. Reports describing its synthesis and receptor activity later appeared in the scientific literature. Commercial availability grew through unregulated channels rather than through pharmaceutical approval.
Melanocortin receptors comprise five subtypes with distinct tissue distributions and functions. Melanotan-2 is described in the literature as a non-selective agonist that engages several of these subtypes, including MC1R, MC3R, MC4R, and MC5R. MC1R is the subtype most directly linked to melanin production in skin cells. Because the compound is not subtype-selective, its observed effects in experimental settings are generally attributed to activity across multiple receptor pathways rather than to a single target.
Melanotan-2 is a synthetic linear peptide built from seven amino acids arranged in a short chain. Its sequence is commonly written as Ac-Nle-Asp-His-D-Phe-Arg-Trp-Lys-NH2, which includes a modified N-terminus and an amidated C-terminus. The molecule belongs to the melanocortin family and acts as a receptor agonist. Structural features such as the D-phenylalanine residue and the Nle substitution are associated with increased stability against enzymatic degradation relative to the natural parent peptide.
Activity is attributed to agonism at melanocortin receptors, particularly MC1R and MC4R. Activation of MC1R on melanocytes increases melanin synthesis, which underlies the reported tanning effect. MC4R engagement in the central nervous system is linked to appetite suppression and to effects on sexual arousal reported in early clinical studies. Those studies were small and were not designed to establish efficacy or long-term safety. Receptor selectivity among the melanocortin subtypes is not absolute, which complicates attribution of any effect to a single pathway.
Regulatory treatment varies between countries. Several national medicines agencies have classified the peptide as unapproved, and customs authorities in some jurisdictions seize shipments on that basis. A few jurisdictions channel supply through prescription-only frameworks that do not list the substance by name. Because the material circulates mainly through online vendors, composition and purity are rarely verified before sale. Surveys of unapproved peptide products have reported labels that did not match measured content in a substantial fraction of samples.
Melanotan II is a synthetic cyclic heptapeptide analogue of alpha-melanocyte-stimulating hormone, a naturally occurring peptide involved in pigmentation signalling. Its structure substitutes a lactam bridge between side chains to increase stability relative to the native hormone. The compound is also known by the shorthand MT-II and by several non-proprietary synonyms used in research catalogues. It is not an approved therapeutic product in any major jurisdiction; material sold under this name is typically offered as a laboratory reagent rather than as a medicine.
=== Eukaryotic sex === Sex in eukaryotes is a composite process, consisting of meiosis and fertilisation, which can be coupled to reproduction. Dacks and Roger proposed on the basis of a phylogenetic analysis that facultative sex was likely present in the common ancestor of all eukaryotes. Early in eukaryotic evolution, about 2 billion years ago, organisms needed a solution to the major problem that oxidative metabolism releases reactive oxygen species that damage the genetic material, DNA. Eukaryotic sex provides a process, homologous recombination during meiosis, for using informational redundancy to repair such DNA damage.
=== Magnetic droplets === The micro-magnetofluidic method is the control of magnetic fluids by an applied magnetic field on a microfluidic platform, offering wireless and programmable control of the magnetic droplets. Hence, the magnetic force can also be used to perform various logical operations, in addition to the hydrodynamic force and the surface tension force. The magnetic field strength, type of the magnetic field (gradient, uniform or rotating), magnetic susceptibility, interfacial tension, flow rates, and flow rate ratios determine the control of the droplets on a micro-magnetofluidic platform. Magnetic droplets, in the context of droplet-based microfluidics, are microliter size droplets that are either composed of ferrofluids or contain some magnetic component that allows for manipulation via an applied magnetic field. Ferrofluids are homogenous mixtures of colloidal solutions of magnetic nanoparticles in a liquid carrier. Two applications of magnetic droplets are the control and manipulation of microfluidic droplets in a microenvironment and the fabrication, transport, and utilization of nanomaterial constructs in the microdroplets. Manipulating magnetic droplets can be used to perform tasks such as arranging droplets into an ordered array for applications in cell culture studies, while the use of magnetic droplets for nanostructure fabrication can be used in drug delivery applications.

=== Announcement === Prompted by the government, the Rhodesian Broadcasting Corporation told the public to stand by for an important announcement from the Prime Minister at 13:15 local time. Smith went first to Government House to inform Gibbs that his Cabinet had declared independence, then to Pockets Hill Studios in east Salisbury to announce UDI to the nation. He read the proclamation aloud, then stated that independence had been declared because it had become "abundantly clear that it is the policy of the British government to play us along with no real intention of arriving at a solution which we could possibly accept ... I promised the people of this country that I would continue to negotiate to the bitter end and that I would leave no stone unturned in my endeavours to secure an honourable and mutually accepted settlement; it now falls to me to tell you that negotiations have come to an end". Smith said that he believed that he would be remiss in his duty if he allowed Rhodesia to continue to "drift in its present paralysing state of uncertainty", and that following Britain's abandonment of the Federation his government was determined that "the same will never be allowed to happen here". He claimed that UDI did not mark "a diminution in the opportunities which our African people have to advance and prosper in Rhodesia", described "racial harmony in Africa" as part of his agenda and condemned black Rhodesian activities as attempts to "blackmail the British government into ... handing the country over to irresponsible rule".

Misuse of the drug has been reported in one case at massive doses of 3,000 to 4,500 mg (100–150 × 30-mg tablets) per day, with the doses gradually increased over time by this individual. No fatalities due to pseudoephedrine misuse have been reported as of 2021. However, death with pseudoephedrine has been reported generally.
Treatments for ATTR-related neuropathy include TTR-specific oligonucleotides in the form of small interfering RNA (patisiran) or antisense inotersen, the former having recently received FDA approval. Research into treatments for ATTR amyloidosis have compared liver transplantation, oral drugs that stabilize the misfolding protein (including tafamidis and diflunisal), and newer therapeutic agents still being investigated (including patisiran). Based on available research, liver transplant remains the most effective treatment option for advanced ATTR amyloidosis, protein stabilizing drugs may slow disease progression but were insufficient to justify delay of liver transplant, and newer agents such as patisiran require additional studies. === Atomic === Lawrencium has three valence electrons: the 5f electrons are in the atomic core. In 1970, it was predicted that the ground-state electron configuration of lawrencium was [Rn]5f146d17s2 (ground state term symbol 2D3/2), per the Aufbau principle and conforming to the [Xe]4f145d16s2 configuration of lawrencium's lighter homolog lutetium. But the next year, calculations were published that questioned this prediction, instead expecting an anomalous [Rn]5f147s27p1 configuration. Though early calculations gave conflicting results, more recent studies and calculations confirm the s2p suggestion. 1974 relativistic calculations concluded that the energy difference between the two configurations was small and that it was uncertain which was the ground state. Later 1995 calculations concluded that the s2p configuration should be energetically favored, because the spherical s and p1/2 orbitals are nearest to the atomic nucleus and thus move quickly enough that their relativistic mass increases significantly. In 1988, a team of scientists led by Eichler calculated that lawrencium's enthalpy of adsorption on metal sources would differ enough depending on its electron configuration that it would be feasible to carry out experiments to exploit this fact to measure lawrencium's electron configuration. The s2p configuration was expected to be more volatile than the s2d configuration, and be more similar to that of the p-block element lead.
== Biosynthesis == The apelin gene encodes a pre-proprotein of 77 amino acids, with a signal peptide in the N-terminal region. After translocation into the endoplasmic reticulum and cleavage of the signal peptide, the proprotein of 55 amino acids may generate several active fragments: a 36 amino acid peptide corresponding to the sequence 42-77 (apelin 36), a 17 amino acid peptide corresponding to the sequence 61-77 (apelin 17) and a 13 amino acid peptide corresponding to the sequence 65-77 (apelin 13). This latter fragment may also undergo a pyroglutamylation at the level of its N-terminal glutamine residue. However the presence and/or the concentrations of those peptides in human plasma has been questioned. Recently, 46 different apelin peptides ranging from apelin 55 (proapelin) to apelin 12 have been identified in bovine colostrum, including C-ter truncated isoforms.
West German SPD leader Oskar Lafontaine was cautious about unification and criticized possible NATO membership for a united Germany, which may have weakened the party's appeal. Meanwhile, Helmut Kohl and the Western CDU made unification the central theme of their campaign, drawing large crowds to rallies in cities such as Erfurt and Chemnitz. The Alliance for Germany organised over 1,400 campaign events and mobilised high-profile Western politicians. In a final blow to the opposition, Democratic Awakening's lead candidate Wolfgang Schnur was exposed by Der Spiegel as a Stasi informant just days before the vote, damaging the credibility of one of the key parties in the alliance.

It is a synthetic cyclic heptapeptide modelled on alpha-melanocyte-stimulating hormone. A lactam bridge links two side chains, forming a ring that stabilises the molecule against proteolysis. It belongs to the broader melanocortin peptide family.
No national medicines agency has approved melanotan-2 for clinical use. Afamelanotide, a related but distinct peptide, holds a marketing authorisation in the European Union for a rare photosensitivity disorder. Melanotan-2 itself is handled as a laboratory chemical.
Its broad activity across melanocortin receptors makes it a tool for probing pigmentation, appetite and vascular signalling. Early trials recorded skin darkening and other effects that were not the original focus of the work. Those observations generated hypotheses that later studies have examined.
It is a synthetic cyclic peptide designed as an analog of alpha-melanocyte-stimulating hormone. It acts on melanocortin receptors and is best known from research into pigmentation. It is not an approved pharmaceutical product.
