== Technology overview == Point-of-care tests are simple medical tests that can be performed at the bedside. In many cases, the simplicity was not achievable until technology developed not only to make a test possible at all but then also to mask its complexity. For example, various kinds of urine test strips have been available for decades, but portable ultrasonography did not reach the stage of being advanced, affordable, and widespread until the 2000s and 2010s. == Adverse effects == Common adverse effects are constipation and nausea, but rates of discontinuation due to constipation were low for both eluxadoline and placebo. Rare adverse effects: fatigue, bronchitis, viral gastroenteritis. Rare serious adverse effects include pancreatitis with a general incidence of 0.3%: higher incidence with 100 mg dose (0.3%) than with 75 mg dose (0.2%). The risk is even greater in those who do not have a gallbladder and the medication is not recommended in this group. In March 2017, the U.S. Food and Drug Administration issued a safety alert for eluxadoline concerning an increased risk of serious pancreatitis in patients without a gallbladder. An FDA review found that in such patients, spasm of the sphincter of Oddi may lead to severe pancreatitis. The FDA reported that in some cases symptoms have occurred with just one or two doses at the recommended dosage for patients without a gallbladder (75 mg). Of two deaths associated with eluxadoline reported up to February 2017, both occurred in patients without a gallbladder.

== History and ideology == The RF was founded on 13 March 1962 in a merger of the Dominion Party (DP), defectors from the anti-Whitehead faction of the United Federal Party (UFP) and former members of the Southern Rhodesia Liberal Party. It was shaky and ideologically split in its early days, with its heterogeneous membership (ranging from advocates of more gradual transition to explicit segregation) united only in their opposition to then-Prime Minister Edgar Whitehead's plans for transition to majority rule, as well as the UK's demands for majority rule before independence. The party harnessed white anxieties of a Congo and Kenya-style majority rule scenario in its successful campaign for the 1962 Southern Rhodesian general election, pledging to keep power "in responsible hands", ensure Southern Rhodesian independence from the Federation, and thwart "this mad idea of a hand-over, of a sell-out of the European and his civilisation, indeed of everything he had put into his country". Its opposition to the UK government's demands for majority rule was so great that the RF-led government eventually declared unilateral independence in 1965. ==== Embryology ==== Cephalopod eggs span a large range of sizes, from 1 to 30 mm in diameter. The fertilised ovum initially divides to produce a disc of germinal cells at one pole, with the yolk remaining at the opposite pole. The germinal disc grows to envelop and eventually absorb the yolk, forming the embryo. == Mechanism == NPRA and NPRB are linked to guanylyl cyclases, while NPRC is G-protein-linked and is a "clearance receptor" that acts to internalise and destroy the ligand. ANP activation of the ANP catalytic receptor will stimulate its intracellular guanylyl cyclase activity to convert GTP to cGMP. cGMP will then stimulate cGMP-dependent protein kinase (PKG), which will then induce smooth muscle relaxation. This is particularly important in the vasculature, where vascular smooth muscle will bind ANP released as a result of increasing right atrial pressure and will cause the walls of the vasculature to relax. This relaxation will decrease total peripheral resistance, which will in turn decrease venous return to the heart. The decrease in venous return to the heart will reduce the preload and will result in the heart's having to do less work. There is also a soluble guanylyl cyclase that cannot be stimulated by ANP. Instead, vascular endothelial cells will use L-arginine to make nitric oxide via nitric oxide synthase. The nitric oxide will then diffuse into the vascular smooth muscle and will activate the soluble guanylyl cyclase. The subsequent increase in cGMP will cause vasodilation with the same effects as described above. This is why nitroglycerine is given to a person having a heart attack. The nitroglycerine will be metabolized to nitric oxide, which will stimulate soluble guanylyl cyclase. This will result in a decrease in total peripheral resistance and a decrease in preload on the heart.
